Losing weight and keeping it off has been the bane of millions of

scientists at the University of Rochester discovered a Master
Hormone called LEPTIN and the dysfunctional message it sends to
the brain that controls appetite and fat storage. Soon after its
discovery, scientists postulated that it is Leptin and -- what is
now simply referred to as -- THE MESSAGE that could determine
if you are fit, or fat!
Leptin, which is produced by fat cells, sends THE MESSAGE to the
brain, telling it whether enough fat is stored in the body and if
it's time to stop eating... But as the human body has evolved,
Leptin has stopped working properly. For most people, this
manifests itself in Leptin sending a continuous, yet erroneous
message to the brain indicating, "your hungry, keep eating, and
store more fat." It is this dysfunctional and perilous message
that results in the body hoarding away even more fat than is
necessary. Simply put, the failure of Leptin and its message is
"sabotaging people's best efforts" at weight loss.
In very simple terms, the higher the leptin levels, the greater
the storage of fat. Correspondingly, if leptin levels can be
lowered, then the body is signaled to release stored and dietary
fat for other purposes, most predominately as energy.
Dr. Philipp E. Scherer may have said it best, when he stated in an
article in the New York Times that:
"The discovery of Leptin established the fat-to-brain
communication axis.”

In view of the impact of Leptin and THE MESSAGE on weight
management, and the link between excess body weight and a host of
pathologies – diabetes, cardiac disease, osteoarthritis, and
metabolic syndrome – the preceding proclamations about Leptin are
not surprising. What is surprising is that finding a solution to
the Leptin conundrum has proved to be very difficult, and neither
continuous exercise nor diets function to lower leptin levels and
fix THE MESSAGE. In fact, popular calorie restricted diets places
the body in a moderate mode of starvation, with the result being
the body becoming more efficient at storing fat and placing the
Leptin messaging to the brain in an even greater state of

To that end, virtually all diets or appetite suppressants rely on
calorie manipulation, a technique that allows for short term
"weight loss" (water and lean muscle loss) and more weight gain

your body to become very efficient at storing fat. Any weight
management measure – most diets and over the counter weight loss
pills – that places the body in a starvation state, will be

DRUG FORMULATIONS IN THE WEIGHT LOSS MARKET
The information outlined in this white paper reviews the current
state of medications available for the treatment of obesity for
comparison to a new product from [Company X]. This product is
called, MaxWLX™, and is a proprietary formulation specifically
developed for the purpose of lowering body fat in individuals
suffering from being overweight or obese. Moreover, the failure of
most dietary supplement products for helping the consumer lose
appreciable body weight and body fat and to offer life time
reductions plus the limitations of current medications to address
obesity as addressed below; offers an excellent opportunity for
MaxWLX™ to achieve success in the US and global distribution
channels.
The leading drug candidates are Rimonanbant, Sibutramine and
Orlistat™ that are currently on the market have different and
varied responses and side effects. As reported by Bray and Ryan;
over a 2 to 4 year period these medications averaged weight loss
of 8%-10% with the placebo contributing 4-6% of that total. Thus,
the net body weight loss is around 4% to 6% with these
medications. The authors underscore that weight maintenance is
maintained as long as the adherence to the medication is
continued.
Rimonabant has the best safety profile of these available drugs.
Nausea and gastrointestinal symptoms are the main reported
findings. Sibutramine elicits a rise in blood pressure and heart
rate that may offer significant downside to a number of patients.
Orlistat™ or its consumer brand, Alli™, induce steatorrhea as the
principal side effect with anal leakage an extenuating factor for
most individuals stopping treatment.
A recent study out of Canada reports the long-term effect of
Orlistat™ and Sibutramine on patient persistence. 17,000
individuals on Orlistat™ and 3500 on Sibutramine were studied (2).
For both Orlistat™ and Sibutramine; only 10% of the individuals
were able to maintain on the medication at 1 year; and a low 2%
after 2 years. The authors concluded that these two drugs offered
very poor long-term persistence rates. One can conclude that these
drugs would offer very limited help for individuals trying to loss
significant amounts of body weight and body fat; and to maintain
those losses over time.
If we are to compare the results of the University of Connecticut
study on the MaxWLX™ blend, a recent study from the University of
Kentucky offers interesting findings that are consistent with
previous literature (3). A multi-center trial was conducted using
a standard dose of Orlistat™ (60 mg; 3 times per day). Overweight
individuals were followed for 16 weeks. The authors reported that
using an intent to treat analysis the Orlistat™ group lost
significantly more weight than placebo (3.05 vs. 1.90 kg). But,
that when those individuals that actually stayed on the medication
for the full 16 weeks the following results were reported (Orlistat™
– 4.8 vs. 3.1 kg; p < 0.001). In comparison, those individuals
consuming MaxWLX™ experienced a reduction in body weight of 9.2 kg
versus 5.4 for the placebo group after only 8 weeks. While
this comparison reflects only one completed study on MaxWLX™ it
does lend support for the belief that MaxWLX™ is a viable option
to these current medications.
The results induced by these medications are modest at best ,and
generally reflect only changes in body weight. There are few, if
any, studies done reporting body composition changes after taking
medications. However, there is one viable study published looking
at body composition changes using Orlistat™. In this study
Orlistat™ was compared to surgical intervention in individuals
(4). After 6 months; both the surgical and Orlistat™ groups had
significant and appreciable reductions in body fat (-10.1 kg
versus -11.4 kg) while at the same maintaining fat free mass or
skeletal tissue. Interestingly, individuals who stayed on the
Orlistat™ treatment for 24 months only experienced a loss of - 4.4
kg compared to the surgical intervention of -15.9 kg. The
subjects in the University of Connecticut study lost -9.8 kg of
body fat after 8 weeks with the placebo losing -5.5 kg. The
subjects in both studies were consuming a moderately restricted
diet while the Connecticut groups were doing matched exercise and
nutritional programming.
As opposed to the side affects noted for most current medications,
the subjects in the University of Connecticut study and
individuals consuming the MaxWLX™ blend for more than one year
have reported very nominal side affects. The most frequent comment
is nominal gastrointestinal distress. However, these side effects
were not substantial enough to require the individuals to come off
the product -- unlike the medications. This provides for an
individual to establish long-term weight loss management.
In summary, the current drug medications available to treat the
global obesity pandemic are few and offer modest success rates. In
addition, these medications have considerable side affects that
make the continued use of said drugs limiting. The safety profile
of the MaxWLX™ blend, and the preliminary efficacy results
obtained from the University of Connecticut study, underscore the
need to move MaxWLX™ into the main stream consumer channel as
rapidly as possible.

INFLUENCES OF A DIETARY SUPPLEMENT IN COMBINATION WITH AN
EXERCISE AND DIET REGIMEN ON ADIPOCYTOKINES AND ADIPOSITY IN WOMEN
WHO ARE OVERWEIGHT

The influence of a high, viscosity polysaccharide dietary
supplement on adipocytokine and regional body composition
responses to a weight loss program was examined. Twenty-two women
(Supplement group (S) (n=11): age= 36.8±7.2 y; weight=87.4 ±6.1
kg; % body fat= 43.6±7.1); Placebo group (P) (n=11): age =38.3±6.8
y; weight = 88.3±7.2 kg; % body fat=44.2±8.2) completed an 8-week
placebo-controlled, double-blind study consisting of a caloric
restricted diet and cardiovascular exercise. Body composition and
serum insulin, leptin, and adiponectin were assessed at pre-,
mid-, and post-intervention.
From pre- to post-intervention, significant decreases (p<0.05) were
observed for body weight (S: 87.4±6.1 to 78.2±3.4 kg; P: 88.3±7.2
to 82.9±4.1 kg) (p<0.05 S vs P), % body fat (S: 43.6±7.1 to
36.2±4.4; P:44.3±8.2 to 40.9±4.3) (p<0.05 S vs P), leptin (S:
28.2±6.1 to 16.3±3.4 μg•ml-1: P: 29.3±9.1 to 19.7±3.3 μg•ml-1)
(p<0.05 S vs P), and insulin (S: 7.2±2.2 to 5.4±2.4 mU•L-1; P:
7.7±2.2 to 5.2±2.7 mU•L-1). Serum adiponectin increased (p<0.05)
(S: 12.2±3.1 to 26.2±3.4 μg•ml-1: 12.6±4.2 to 21.7±4.3 μg•ml-1)
(p<0.05 for S vs P).
Data indicated the
dietary supplement used during an 8-week weight loss/exercise
program exhibited favorable effects on adipocytokines and regional
body composition.
